I think Steve Austins Title win at WrestleMania 14 was one of the biggest, as it made Steve Austin the most popular WWE Superstar of all time.
I also think Chris Benoits was a big one at WrestleMania 20 but I think that was because it was the most emotional Title win in WrestleMania history.

I don't think you can compare title wins pre-brand split with those that came after, because winning a championship in the post-brand split WWE just doesn't mean as much with titles and rosters being as watered down as they are.

I presume you mean world title. And by the way Benoits World Heavyweight Championship win was not his first world title he won the WCW title once.
Just in the WWE otherwise Big Show/Ric Flair/Booker T...Ect,Ect would not count. As their first Title reigns was outside the WWE.

Eddies first title win and the celebration will always stick out for me, absolutely great moment
Benoit's a month later at Mania was another really great moment
Edge's first title win cashing in MITB was also very memorable just because it was so new and fresh

Undertaker over Hogan at Survivor Series 1991
"The boyhood dream has come true" at WrestleMania 12
"The Austin Era has begun" at WrestleMania 14
The Rock becoming the "Corporate Champion" at Survivor Series 1998
Foley over The Rock on Raw 1999
Brock becoming the youngest WWE Champion ever over The Rock who was the youngest before him at SummerSlam 2002
Eddie over Brock at No Way Out 2004
Edge cashing in Money in the Bank on John Cena at New Year's Revolution 2006

No Christian?
That was a great moment too but those moments immediately came to mind when I was posting. I'd also add Goldberg over Hogan on Nitro in '98, DDP in the 4-way with Hogan/Sting/Flair at Spring Stampede '99, and Ron Simmons over Vader in '92.

Batista. He had a goldberg like push from RR all the way until Mania and IWC expected Triple H to win at WM 21. Batista winning was a great moment and a game changer.

Shawn Michaels in '96
Stone Cold in '98

Both of these guys were the most over guy in the WWF well before their first title win, and each of them was continuously built up until the point of winning it. This was also back when the IC Title was very meaningful, so they each used that belt as a proper stepping stone. They each won their titles in the classic Wrestlemania Main Event passing-of-the-torch fashion (hell of a match each time too). Each of these guys had such a memorable road to the WWF Title as well as a memorable match for the WWF title.
