Being a good friend in this hypothetical would probably entail you strongly suggesting to your friend to work out his relationship with the primary woman since "cheating" tends to create emotional complications which can become ugly in a hurry, not secretly rat him out to someone who is of lesser personal importance to you than he is, seeing as he's ostensibly your "best mate" according to your post... Although honestly, OP, your post is so muddled I'm not sure what the chronology of events is. If the woman he cheated on is his "ex" what does it matter now anyway? Which girl was dumped? Why did he dump someone because he was diagnosed with depression? You're the friend of the girl with whom he is "cheating," or...? You relied too heavily on personal pronouns for me to make heads or tails of what you're saying, precisely.
